On August 28, GlaxoSmithKline (GSK) announced that its new oral antibiotic Gepotidacin has been approved by the UK Medicines and Healthcare products Regulatory Agency (MHRA) for the treatment of uncomplicated urinary tract infections (uUTIs).
This is the first new oral antibiotic in nearly 30 years approved for this condition — a major milestone in the fight against bacterial infections.
Earlier this year, in March, the U.S. FDA had already given the green light to Gepotidacin.
According to the MHRA, results from two latestage clinical trials showed that Gepotidacin is at least as effective as current standard treatments, and in some cases, even more effective.

A Unique Mechanism That Reduces Resistance
Gepotidacin is the first drug of a brand-new class called triazaacenaphthylene antibiotics.
Unlike traditional antibiotics, it works through a dual mechanism: blocking both DNA gyrase and topoisomerase IV, two enzymes bacteria need to replicate their DNA.Because bacteria would need to mutate in both enzymes to become resistant, the risk of resistance developing is much lower compared to older drugs.

Strong Clinical Trial Results
Two pivotal Phase III trials (EAGLE-2 and EAGLE-3) enrolled more than 3,100 patients and directly compared Gepotidacin (1500mg, twice daily) with the current standard drug nitrofurantoin (100mg, twice daily).
•        EAGLE-2: Success rate 50.6% vs. 47% (nitrofurantoin)
•        EAGLE-3: Success rate 58.5% vs. 43.6% (nitrofurantoin)
In both studies, Gepotidacin performed better overall.

Beyond UTIs: Promising Results in Gonorrhea
Gepotidacin is also showing strong potential against drug-resistant gonorrhea.
In the EAGLE-1 trial, published in The Lancet in April 2025, it achieved a 92.6% cure rate, comparable to current injection-based treatments.
Based on this data, the FDA accepted a supplemental application in August 2025 to review Gepotidacin for treating uncomplicated urogenital gonorrhea, granting it priority review with a decision expected by December 11, 2025.
If approved, it would be the first new antibiotic for gonorrhea in 30 years with an entirely new way of working.

Market Outlook: A Core Launch for GSK
GSK has highlighted antibiotics as a strategic focus. In the first half of 2025, the company reported £15.5 billion ($20.2 billion) in revenue, up 5% year-on-year, with specialty medicines leading the growth.
Gepotidacin is one of GSK’s five key launches for 2025, with peak sales expected to exceed £2 billion.
Goldman Sachs, however, takes a more conservative view, forecasting sales of £70 million in 2025 and £681 million by 2035.

What It Means for China
In China, progress on new antibiotics for resistant infections has been relatively slow. It’s highly likely that Gepotidacin will eventually enter the market through import or licensing.
